Manufacturing Quality Control of HEPES

Appearance Visual Inspection Take an appropriate amount of sample and place it on a white background. Observe its color and appearance under natural light or standard light sources. It should appear as a white to off-white crystalline powder.
Identification Infrared Spectroscopy (IR) Prepare a KBr pellet of the sample and measure its infrared absorption spectrum. Compare it with the reference spectrum of the standard substance. The main absorption peak positions should be consistent.
Identification Nuclear Magnetic Resonance Hydrogen Spectroscopy (¹H NMR) Dissolve the sample in deuterated water (D₂O) or another suitable deuterated solvent, and measure the ¹H NMR spectrum. The characteristic chemical shift should match the standard spectrum.
pH Value pH Meter Method Prepare a solution of a certain concentration (e.g., 1% w/v) in water. Measure the pH value using a calibrated pH meter at a specified temperature (usually 25°C). The pH value should fall within the specified range (e.g., 2.8–3.2).
Content Determination HPLC (High-Performance Liquid Chromatography) Use a reverse-phase C18 column with a mobile phase consisting of phosphate buffer-ethanol, etc. Detect using a UV detector (e.g., 210 nm). Calculate the content of the main component using the external standard method, typically requiring ≥99.0%.
Content Determination Acid-Base Titration Dissolve the sample in water and titrate with a standard sodium hydroxide solution. Determine the endpoint using potentiometry or an indicator. Calculate the HEPES content. This method is suitable for rough quantification when there are no interfering impurities.
Dry Loss Thermogravimetric Analysis (TGA) or Atmospheric Drying Dry the sample at 105°C until constant weight is reached. Calculate the moisture and volatile matter content based on the difference in mass before and after drying. Typically, this should be ≤0.5%.
Residual Ash High-Temperature Incineration Char the sample and incinerate it in a high-temperature furnace (700–800°C) until constant weight is achieved. Weigh the residual inorganic matter and calculate its mass. Typically, this should be ≤0.1%.
Heavy Metals Thioacetamide Method (or ICP-MS) Follow the pharmacopoeia method to process the sample, add thioacetamide for coloring, and determine by colorimetry. Alternatively, use Inductively Coupled Plasma Mass Spectrometry (ICP-MS) for trace metal element quantification. Total heavy metals are usually required to be ≤10 ppm.
Chlorides Silver Nitrate Turbidimetry Acidify the sample solution with nitric acid and add silver nitrate. Compare the turbidity with that produced by a standard sodium chloride solution to control the level of chloride impurities.
Sulfates Barium Chloride Turbidimetry Acidify the sample solution with hydrochloric acid and add barium chloride. Compare the turbidity with that produced by a standard potassium sulfate solution to control sulfate impurities.
Related Substances (Impurities) HPLC (High-Performance Liquid Chromatography) Use gradient elution or isocratic conditions to separate and detect potential organic impurities (such as synthetic intermediates or degradation products). Individual impurities should typically be ≤0.1%, and total impurities ≤0.5%.
Moisture Karl Fischer Titration Determine the moisture content in the sample using either the volumetric or coulometric Karl Fischer reagent. This method provides precise measurement of water content, typically requiring ≤0.5%.
